Objective: Theory and research suggest that the pursuit of personal goals that do not fit a person's affect-based implicit motives results in impaired emotional well-being, including increased symptoms of depression. The aim of this study was to evaluate an intervention designed to enhance motive-goal congruence and study its impact on well-being. Method: Seventy-four German students (mean age = 22.91, SD = 3.68; 64.9% female) without current psychopathology, randomly allocated to 3 groups: motivational feedback (FB; n = 25; participants learned about (...) the fit between their implicit motives and explicit goals), FB + congruence-enhancement training (CET; n = 22; participants also engaged in exercises to increase the fit between their implicit motives and goals), and a no-intervention control group (n = 27), were administered measures of implicit motives, personal goal commitments, happiness, depressive symptoms, and life satisfaction 3 weeks before (T1) and 6 weeks after (T2) treatment. Results: On two types of congruence measures derived from motive and goal assessments, treated participants showed increases in agentic (power and achievement) congruence, with improvements being most consistent in the FB+CET group. Treated participants also showed a trend-level depressive symptom reduction, but no changes on other well-being measures. Although increases in overall and agentic motivational congruence were associated with increases in affective well-being, treatment-based reduction of depressive symptoms was not mediated by treatment-based agentic congruence changes. Conclusion: These findings document that motivational congruence can be effectively enhanced, that changes in motivational congruence are associated with changes in affective well-being, and they suggest that individuals’ implicit motives should be considered when personal goals are discussed in the therapeutic process. We investigated how two cues to contrast—beat gesture and contrastive pitch accenting—affect comprehenders' cognitive load during processing of spoken referring expressions. In two visual‐world experiments, we orthogonally manipulated the presence of these cues and their felicity, or fit, with the local (sentence‐level) referential context in critical referring expressions while comprehenders' task‐evoked pupillary responses (TEPRs) were examined. In Experiment 1, beat gesture and contrastive accenting always matched the referential context of filler referring expressions and were therefore relatively felicitous on the global (...) (experiment) level, whereas in Experiment 2, beat gesture and contrastive accenting never fit the referential context of filler referring expressions and were therefore infelicitous on the global level. The results revealed that both beat gesture and contrastive accenting increased comprehenders' cognitive load. For beat gesture, this increase in cognitive load was driven by both local and global infelicity. For contrastive accenting, this increase in cognitive load was unaffected when cues were globally felicitous but exacerbated when cues were globally infelicitous. Together, these results suggest that comprehenders' cognitive resources are taxed by processing infelicitous use of beat gesture and contrastive accenting to convey contrast on both the local and global levels. Two strains of male mice have bred over fortygenerations, starting with the work of RobertCairns and his colleagues, one strain with ahigh level of intra-species aggression, theother a low level of aggression. Thehigh-aggression mice tend to establishdominance hierarchies and particularly fight inthe presence of female mice. Thelow-aggression mice tend, in groups of theirown, to have a high degree of low-intensity,peaceful social contact, and to be more timidin initiating action than the high-aggressionmice. Biochemical differences have beenobserved between the two strains, and (...) confirmedby the present data: the high-aggression micehave greater dopamine concentrations (in thecaudate nucleus and nucleus accumbens), lowerlevels of the stress hormone corticosterone,and higher levels of testosterone than thelow-aggression mice. Finally, what are thesocial and neurochemical mechanisms by whichhigh-aggression mice can develop low-aggressionbehavior if brought out of isolation and intogroups?Maternal separation was found to lead todecreases in stress levels, as measured bycorticosterone, in the low-aggression but notthe high-aggression, mice – presumably becauseof the surplus of maternal care the pupsreceive on returning to the nest. When alow-aggression mouse became dominant and ahigh-aggression mouse became submissive, theirusual pattern of corticosterone andtestosterone levels was found to be reversed. The change to low-aggression behavior inhigh-aggression mice switching from anisolation condition to a group condition, wasmediated by a decrease in D 1 dopaminereceptor densities.These results, like the ones on which theybuild, argue for substantial developmentalinfluences in expressions of the genesinfluencing aggressive or cooperative behavior. When repeated requests for heat fell on the pastor's deaf ears, the sisters requested to leave the parish and were told that they would then also have to leave another thriving ministry in the diocese. Fifty years later in Miami, Florida, when another conflict arose, the sisters involved discerned a very different response to injustice, one marked by concern for "justice, honesty, and personal responsibility." The model of a "good sister" had shifted from an emphasis on obedience and institutional loyalty toward shared responsibility for ministry and leadership.Chapter 2 surveys the landscape of women's engagement with the Church and its structures of leadership and adapts H. Richard Niebuhr's Christ and Culture to sketch five "types": women against the Church; women content with the Church; women above the Church; women and Church in paradox; and women transforming Church. Background Payment of research participants helps to increase recruitment for research studies, but can pose ethical dilemmas. Research ethics committees (RECs) have a centrally important role in guiding this practice, but standardisation of the ethical approval process in Ireland is lacking. Aim Our aim was to examine REC policies, experiences and concerns with respect to the payment of participants in research projects in Ireland. Method Postal survey of all RECs in Ireland. Results Response rate was 62.5% (n=50). 80% of RECs (...) reported not to have any established policy on the payment of research subjects while 20% had refused ethics approval to studies because the investigators proposed to pay research participants. The most commonly cited concerns were the potential for inducement and undermining of voluntary consent. Conclusions There is considerable variability among RECs on the payment of research participants and a lack of clear consensus guidelines on the subject. The development of standardised guidelines on the payment of research subjects may enhance recruitment of research participants. (shrink)

Transmissible cancers are elusive and understudied parasitic life forms caused by malignant clonal cells (nine lineages are known so far). They emerge by completing sequential steps that include breaking cell cooperation, evade anti‐cancer defences and shedding cells to infect new hosts. Transmissible cancers impair host fitness, and their importance as selective force is likely largely underestimated. It is, therefore, crucial to determine how common they might be in the wild. Here, we draw a parallel between the steps required for a (...) transmissible cancer to emerge and the steps required for an intelligent civilisation to emerge in the Milky Way using a modified Drake equation. Using numerical analyses, we estimate the potential number of extant marine and bivalve species in which transmissible cancers might exist. Our results suggest that transmissible cancers are more common than expected, and that new lineages can be found by screening a large number of species. (shrink)

Plasmodium falciparum is the most deadly human malarial parasite, responsible for an estimated 207 million cases of disease and 627,000 deaths in 2012. Recent studies reveal that the parasite actively regulates a large fraction of its genes throughout its replicative cycle inside human red blood cells and that epigenetics plays an important role in this precise gene regulation. Here, we discuss recent advances in our understanding of three aspects of epigenetic regulation in P. falciparum: changes in histone modifications, nucleosome occupancy (...) and the three‐dimensional genome structure. We compare these three aspects of the P. falciparum epigenome to those of other eukaryotes, and show that large‐scale compartmentalization is particularly important in determining histone decomposition and gene regulation in P. falciparum. We conclude by presenting a gene regulation model for P. falciparum that combines the described epigenetic factors, and by discussing the implications of this model for the future of malaria research. Biological ontologies are used to organize, curate, and interpret the vast quantities of data arising from biological experiments. While this works well when using a single ontology, integrating multiple ontologies can be problematic, as they are developed independently, which can lead to incompatibilities. The Open Biological and Biomedical Ontologies Foundry was created to address this by facilitating the development, harmonization, application, and sharing of ontologies, guided by a set of overarching principles. One challenge in reaching these goals was that the (...) OBO principles were not originally encoded in a precise fashion, and interpretation was subjective. Here we show how we have addressed this by formally encoding the OBO principles as operational rules and implementing a suite of automated validation checks and a dashboard for objectively evaluating each ontology’s compliance with each principle. This entailed a substantial effort to curate metadata across all ontologies and to coordinate with individual stakeholders. We have applied these checks across the full OBO suite of ontologies, revealing areas where individual ontologies require changes to conform to our principles. Our work demonstrates how a sizable federated community can be organized and evaluated on objective criteria that help improve overall quality and interoperability, which is vital for the sustenance of the OBO project and towards the overall goals of making data FAIR. Bayesian confirmation theory is rife with confirmation measures. Zalabardo focuses on the probability difference measure, the probability ratio measure, the likelihood difference measure, and the likelihood ratio measure. He argues that the likelihood ratio measure is adequate, but each of the other three measures is not. He argues for this by setting out three adequacy conditions on confirmation measures and arguing in effect that all of them are met by the likelihood ratio measure but not by any of the other (...) three measures. Glass and McCartney, hereafter “G&M,” accept the conclusion of Zalabardo’s argument along with each of the premises in it. They nonetheless try to improve on Zalabardo’s argument by replacing his third adequacy condition with a weaker condition. They do this because of a worry to the effect that Zalabardo’s third adequacy condition runs counter to the idea behind his first adequacy condition. G&M have in mind confirmation in the sense of increase in probability: the degree to which E confirms H is a matter of the degree to which E increases H’s probability. I call this sense of confirmation “IP.” I set out four ways of precisifying IP. I call them “IP1,” “IP2,” “IP3,” and “IP4.” Each of them is based on the assumption that the degree to which E increases H’s probability is a matter of the distance between p and a certain other probability involving H. Knowledge of human uses of animals is an important, but understudied, aspect of how humans treat animals. We developed a measure of one kind of knowledge of human uses of animals – knowledge of factory farming. Studies 1 (N = 270) and 2 (N = 270) tested an initial battery of objective, true or false statements about factory farming using Item Response Theory. Studies 3 (N = 241) and 4 (N = 278) provided evidence that responses to a 10-item Knowledge (...) of Factory Farming Scale predicted a reduction in consumption of animal products (rs = −.17- −.27) and approval of political actions aimed at factory farming (rs = .2 – .24). Path models from Studies 3 and 4 suggested that different kinds of knowledge uniquely predicted different outcomes. The Knowledge of Factory Farming scale was a unique predictor of approval of political actions concerning factory farmed animals but not animal consumption. Knowledge of Animals Used as Food predicted animal consumption but not political actions concerning farmed animals. These results highlight that different kinds of knowledge can be relevant for different animal related outcomes. (shrink)

Airway mucin secretion is important pathophysiologically and as a model of polarized epithelial regulated exocytosis. We find the trafficking protein, SNAP23, selectively expressed in secretory cells compared with ciliated and basal cells of airway epithelium by immunohistochemistry and FACS, suggesting that SNAP23 functions in regulated but not constitutive epithelial secretion. Heterozygous SNAP23 deletant mutant mice show spontaneous accumulation of intracellular mucin, indicating a defect in baseline secretion. However mucins are released from perfused tracheas of mutant and wild-type mice at the (...) same rate, suggesting that increased intracellular stores balance reduced release efficiency to yield a fully compensated baseline steady state. In contrast, acute stimulated release of intracellular mucin from mutant mice is impaired whether measured by a static imaging assay 5 min after exposure to the secretagogue ATP or by kinetic analysis of mucins released from perfused tracheas during the first 10 min of ATP exposure. Together, these data indicate that increased intracellular stores cannot fully compensate for the defect in release efficiency during intense stimulation. The lungs of mutant mice develop normally and clear bacteria and instilled polystyrene beads comparable to WT mice, consistent with these functions depending on baseline secretion that is fully compensated. Cette serie est destinee a regrouper des etudes thematiques faisant le point sur differents aspects de l'histoire ou de la culture syriaques, celles des communautes chretiennes dont la langue de culture est le syriaque (maronites, syriaques catholiques et orthodoxes, assyro-chaldeens, communautes du Proche-Orient et de l'Inde...). Cible de jugements malveillants pendant un siecle, la philosophie en syriaque a recemment ete l'objet d'un renouveau d'interet qui en a fait l'un des sujets les plus debattus dans les etudes syriaques. Les Syriaques ont (...) ete regardes dans le meilleur des cas comme de simples transmetteurs aux musulmans de la philosophie grecque, incapables d'une pensee philosophique originale. Si leur role crucial de vecteurs culturels n'est pas en question, ce volume se propose pourtant de montrer aussi la specificite des formes que la philosophie grecque a revetues en monde syriaque au long d'un millenaire, du iie au xiiie siecle. Des le debut, la selection des themes et textes philosophiques par les Syriaques temoigne d'une continuite avec les courants contemporains de l'Empire romain et d'une participation active aux debats qui y avaient cours. Un auteur syriaque comme Bardesane pouvait s'inserer avec originalite dans les controverses du monde imperial sur le determinisme et la providence. La traduction de textes grecs d'ethique et l'utilisation de textes de sagesse attribues aux philosophes grecs attestent un interet pour la philosophie comme " exercice spirituel ", partage par les auteurs chretiens de langue grecque et de langue syriaque, qui dans les memes annees s'interrogeaient sur la valeur de la paideia hellenique pour l'education du chretien. On pourrait donc parler pour ces premiers siecles d'une philosophie chretienne greco-syriaque. A partir du vie siecle, surtout par l'intermediaire des miaphysites, la logique aristotelicienne penetre en monde syriaque. La figure cle pour ce passage est Sergius de Res'ayna (m. 536), et encore une fois la continuite avec le monde grec marque cette entreprise : les commentaires aux uvres logiques d'Aristote ecrits par Sergius et ses successeurs, en particulier au monastere de Qennere, reprennent la structure des commentaires grecs produits a Alexandrie dans l'Antiquite tardive. Des le viiie siecle on assiste aussi au developpement de la logique parmi les elites ecclesiastiques syro-orientales. L'essor de la falsafa, la philosophie d'empreinte grecque en monde musulman, produit une rupture de cette continuite greco-syriaque. Les Syriaques, decisifs dans la transmission de la philosophie a l'Islam, ressentent de plus en plus l'influence de la falsafa. A la fin du premier millenaire, des auteurs chretiens de culture syriaque ecrivent des ouvrages philosophiques en arabe et participent a la phase formative de la falsafa ; au debut du deuxieme millenaire paraissent des ouvrages philosophiques qui, ecrits en syriaque, refletent les structures conceptuelles de la falsafa. C'est la naissance d'une philosophie syro-arabe, qui culmine dans l'uvre de Barhebraeus (xiiie siecle). Le volume s'enrichit de deux contributions bibliographiques qui fourniront au lecteur un panorama exhaustif des sources et des etudes qui ont ete consacrees a la philosophie en syriaque. (shrink)

The Plasmodium falciparum genome‐sequencing project has provided malariologists with vast amounts of new information pertinent to a multitude of cellular processes that previously were only guessed about. In exploring this morass of predicted genes and proteins, there is now a danger of simply re‐inventing the cell. Fortunately, new global transcriptional analyses reassure malariologists that they are not dealing with just “any old cell.” The informative papers on the plasmodial transcriptome by Le Roch et al. (2003)1 and Bozdech et al. (...) (2003)2 discussed below forge a bridge between the genomics and proteomics of P. falciparum. Institutional Review Board decisions hinge on the availability and interpretation of information. This is demonstrated by the following well-known historical example. In 2001, 24-year-old Ellen Roche died from respiratory distress and organ failure as a result of her participation in a study at Johns Hopkins Asthma and Allergy Center. The non-therapeutic physiological study, “Mechanisms of Deep Inspiration-Induced Airway Relaxation,” was designed to examine airway hyperresponsiveness in healthy individuals in order to better understand the pathophysiology of asthma. Participants inhaled hexamethonium, a (...) chemical ganglionic blocker that was not FDA-approved for use as specified in the experimental protocol. Risks of inhaling hexamethonium, including lung damage, had been reported in the 1950s and ‘60s. An investigation conducted after Roche's death determined that the study's principal investigator had failed to adequately search the medical literature; the protocol submitted for IRB review cited articles indexed in online literature databases that went back only as far as the 1970s. Is God's foreknowledge compatible with human freedom? One of the most attractive attempts to reconcile the two is the Ockhamistic view, which subscribes not only to human freedom and divine omniscience, but retains our most fundamental intuitions concerning God and time: that the past is immutable, that God exists and acts in time, and that there is no backward causation. In order to achieve all that, Ockhamists distinguish ‘hard facts’ about the past which cannot possibly be altered from ‘soft facts’ (...) about the past which are alterable, and argue that God's prior beliefs about human actions are soft facts about the past. With help from the recent work of John Wall, Mary M. Doyle Roche, Bonnie J. Miller-McLemore, and others, the author claims children's right to work in Ethna Regan's sense, as an expression of a "situated universal." Rights on this view are real but contingent. They (...) are means to protect universal human goods and vulnerabilities under attack in particular situations. In this case, a situated right to work protects children's flourishing in a global culture still shaped by liberal and capitalist institutions, even as it criticizes that culture's injustices and exclusions.
